Regulation of substrate recruitment by the Ubiquitin-mediated proteasome pathway

The irreversibility of ubiquitin(Ub)-mediated proteolysis makes it the ultimate on-off switch of a cell. In contrast to many protein-protein post-translational modifiers/modification, Ub/ubiquitination and the 26S proteasome-mediated degradation (UPS) enables the regulation and control of a complex and diverse array of biological processes. The complexity in its process rivals that of transcription and is often described as the nexus of plant biology. While the proteasome activity is regulated at several levels, substrate recruitment by the E3 ligases is an increasingly engaging area of research. Special mention goes to the SCF E3 ligases that are the most abundant in both the plant and animal kingdoms; and in the last two decades since the first report, much have been known about the roles of F-box proteins in the regulation of important biological processes. However, a lot more needs to be discovered to understand the mechanisms of the entire process of the UPS and harness the knowledge for various translational and therapeutic studies.
